Abstract:
Metabolic pathway in brain is important but challenging for traditional methodology based upon combination of liquid chromatography and mass spectrometry. We have developed in-situ mass spectrometry methods for direct metabolites measurements in both large scale tissue slices and single live cells.
To achieve above goals, our group has developed novel ambient ionization devices including desorption electrospray ionization and induced electrospray ionization, which overcome severe ion suppression effect from biological samples. Multiples metabolites were directly detected in brain regions on brain slices and single living neurons on fresh isolated brain tissues. Mass spectrometry imaging enabled direct visualization of multiple metabolic pathway disorder in AD progress. Single cell mass spectrometry enabled metabolites identifications and discovery of novel metabolic pathway in single living neurons.
