Abstract:
Splicing dysregulation is one of the molecular hallmarks of cancer. However, the underlying molecular mechanisms remain poorly defined. Here we report that the splicing factor RBM4 suppresses proliferation and migration of various cancer cells by specifically controlling cancer-related splicing. Particularly, RBM4 regulates Bcl-x splicing to induce apoptosis, and coexpression of Bcl-xL partially reverses the RBM4-mediated tumor suppression. Moreover, RBM4 antagonizes an oncogenic splicing factor, SRSF1, to inhibit mTOR activation. Strikingly, RBM4 expression is decreased dramatically in cancer patients, and the RBM4 level correlates positively with improved survival. In addition to providing mechanistic insights of cancer-related splicing dysregulation, this study establishes RBM4 as a tumor suppressor with therapeutic potential and clinical values as a prognostic factor.
Introduction:
Yang obtained his Ph.D. in Cell Biology from the State Key Laboratory of Molecular Oncology in the Cancer Institute of Peking Union Medical College of Tsinghua University in 2007. He was supervised by Prof. Qimin Zhan. Subsequently,Yang did his postdoctoral research in the Lineberger Cancer Center at the University of North Carolina at Chapel Hill from 2007 to 2011. At the end of 2011, Yang was promoted to a Research Assistant Professor, and supported by the UNC Research Council. In addition, he was awarded the “Young Investigator Development award” by UNC. In 2013, Yang came back to Dalian Medical University to work as a full-time professor. In 2014, Yang was supported by the NSFC Excellent Young Scientists grant, and awarded the Newton Advanced Fellowship from the Royal Society and the Academy of Medical Sciences, UK. In 2016, he obtained the Distinguished Young Scientist award from Dalian government. Moreover, as the second awardee, Yang obtained the Science and Technology Award, and the Natural Science Award of Liaoning province in 2017. Additionally, he was supported by the NSFC Key project in 2018. Currently, Yang has published more than 30 scientific papers in the international prestigious journals (total five-year impact factor: 283.8), including as the first or corresponding author publised15 papers in Nature Methods, Nature Structural & Molecular Biology (2), Cancer Cells, and Nature Communications, which had been cited 740 times in the most recent five years.
