Lecture: Development and Application of Synthetic Biology Tools in Saccharomyces cerevisiae

Abstract:

Metabolic engineering involves three important steps: modification of the cellular pathway (synthesis), rigorous evaluation of the phenotype of the modified cell (phenotypic characterization), and the design of further modifications (optimal design) based on the results of the evaluation. For the complexity of biological systems, many rounds of metabolic engineering cycle are necessary to engineered strains.

Synthetic biology technology has greatly accelerate the strain construction process. Compared with the plasmid expression, integrating of the gene on the genome ensure stable gene expression for a long time. The speaker proposed the integration of the target DNA fragment into the delta sites of the multi-copy of the yeast genome, and screened the strain integrated multiple target DNA fragments by rounds of stress resistance. Subsequently, in order to avoid using resistance, A CRISPR technique was developed to integrate multiple copies of the metabolic pathway without any trace into the yeast genomic Delta site in one step. This precise and efficient method enables the simultaneous editing of multiple sites on the genome with large DNA fragments inserted.

Introduction:

SHI Shuobo, Ph.D., works as a Professor in Beijing University of Chemical Technology. He received his Ph.D. degree from Tianjin University in 2009. From 2009 to 2012 he worked as a postdoctoral in Jens Nielsen lab at Chalmers University of Technology, Sweden. His research interests focus on fundamental and applied research in metabolic engineering, systems biology and synthetic biology, building and engineering microbes for efficient production of biofuels, chemicals and other biological products. Representative work was published in high-level journals such as Metab. Eng., MBio, Biotechnol. Biofuels with a total citation of more than 500 times.
